Scientists Discover the Link Between Deep Sleep and Growth Hormone

A good night’s sleep not only helps the body relax and recover but also plays a crucial role in regulating the release of growth hormone. This hormone is essential for building muscles and bones, burning fat, and supporting healthy growth and development. According to a report by ScienceDaily, citing a study published in the journal Cell, athletes rely on quality sleep for physical recovery, while young people need sufficient sleep for optimal growth and development.

Scientists have long known that growth hormone release peaks during sleep, particularly during non-rapid eye movement (NREM) or deep sleep. However, the neural mechanism controlling this process had remained a mystery until now.

Researchers at the University of California have identified the neural circuits responsible for regulating growth hormone release during sleep. They also discovered a previously unknown feedback system that helps maintain balanced levels of the hormone within the body.

This breakthrough provides new insights into the complex relationship between sleep and hormonal regulation. It could pave the way for innovative treatments for sleep disorders associated with metabolic diseases such as diabetes, neurodegenerative disorders like Parkinson’s and Alzheimer’s, obesity, and cardiovascular disease, as growth hormone plays a key role in regulating glucose and fat metabolism.

Growth Hormone and the Brain

Professor Shenghao Ding, a neuroscientist at the University of California and the study’s lead researcher, explained that the nerve cells responsible for coordinating growth hormone release are located deep within the hypothalamus. These include two distinct types of neurons: those that release growth hormone-releasing hormone (GHRH) and those that release somatostatin.

Once growth hormone is released, it activates neurons in the locus coeruleus, a region in the brainstem responsible for wakefulness, attention, thinking, and responding to new experiences. Dysfunction in the locus coeruleus has been linked to several neurological and psychiatric disorders.

Understanding the Locus Coeruleus

Co-author Daniel Silverman noted that understanding the neural circuit responsible for growth hormone release could eventually lead to new hormone-based therapies aimed at improving sleep quality or restoring the body’s natural hormonal balance. He added that some experimental gene therapies already target specific cells, and this neural circuit may offer a new way to suppress excessive activity in the locus coeruleus.

Advanced Research Techniques

Using advanced neural circuit-tracing techniques, the research team discovered that the two peptide hormones regulating growth hormone release behave differently depending on the stage of sleep.

During rapid eye movement (REM) sleep, levels of both GHRH and somatostatin increase, leading to greater growth hormone release. During NREM sleep, somatostatin levels decline while GHRH rises only slightly, creating a distinct pattern of hormonal regulation.

The researchers also identified another previously unknown feedback mechanism involving the locus coeruleus. As growth hormone gradually accumulates during sleep, it stimulates the locus coeruleus, encouraging wakefulness. However, if activity in the locus coeruleus becomes excessively high, it unexpectedly begins to promote drowsiness instead.

A Delicate Balance Between Sleep and Growth Hormone

Silverman explained that the findings reveal a finely balanced system between sleep and growth hormone.

“Sleep deprivation reduces the release of growth hormone. Rising levels of growth hormone push the brain toward wakefulness. Sleep stimulates the release of growth hormone, which in turn regulates wakefulness. This balance is essential for growth, tissue repair, and metabolic health.”

Given the effects of growth hormone on the locus coeruleus, the newly discovered system may also influence attention and other cognitive functions. This suggests that growth hormone is involved in far more than simply building muscles and bones—it may play an important role in maintaining overall brain function and human health.
